Early non-invasive diagnosis is required to increase the effectiveness of anticancer therapy.</p><p><bold>Objective: </bold>to determine the content of circulating tumor cells (CTCs) and their derivatives in the peripheral blood using the MDA-231 aptamer, compare the results obtained with the clinical and molecular characteristics of BC.</p><p><bold>Materials and methods.</bold> The study included 22 patients with BC. Detection of CTCs and circulating tumor microemboli was carried out in 3.5 ml of the blood of BC patients with the help of the MDA-231 aptamer which is affine for breast cancer cells, labeled with the fluorescent Cy3 dye. The count of CTCs in the blood samples was performed using fluorescent and laser scanning microscopy.</p><p><bold>Results and conclusions. </bold>The content of CTCs and circulating tumor microemboli in the peripheral blood of patients with BC of various molecular subtypes was analyzed using the MDA-231 aptamer. The relationship between the number of CTCs and the molecular biological subtype was revealed. The obtained results show the possible prognostic value of CTCs use for monitoring effectiveness of anticancer therapy and control of recurrence of BC.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение. </bold>Рак молочной железы (РМЖ) – наиболее распространенный вид злокачественных новообразований среди женщин, имеющий высокий уровень метастазирования. Для увеличения эффективности противоопухолевой терапии необходима ранняя неинвазивная диагностика.</p><p><bold>Цель исследования</bold> – определить содержание циркулирующих опухолевых клеток (ЦОК) и их производных в периферической крови с помощью аптамера MDA-231, полученные результаты сравнить с клинико-молекулярными характеристиками РМЖ.</p><p><bold>Материалы и методы. </bold>В исследование включены 22 пациентки с РМЖ. Детекцию ЦОК и циркулирующих опухолевых микроэмболов проводили в 3,5 мл крови больных РМЖ с помощью аффинного клеткам РМЖ аптамера MDA-231, меченного флуоресцентным красителем Cy3. Подсчет ЦОК в мазках образцов крови проводили с помощью флуоресцентной и сканирующей лазерной микроскопии.</p><p><bold>Результаты и выводы. </bold>C помощью аптамера MDA-231 проанализировано содержание ЦОК и циркулирующих опухолевых микроэмболов в периферической крови больных РМЖ различных молекулярных подтипов. Выявлена зависимость между количеством ЦОК и молекулярно-биологическим подтипом РМЖ.
